ABSTRACT

The COVID-19 pandemic presented new challenges for scholars and government officials to predict people's evacuation decisions under a conflicting natural disaster. In this study, we examined households' evacuation and shelter intentions given the potential conflicts between the perceived risks from a hurricane and the coexisting public health crisis. We surveyed households living inside hurricane evacuation zones in Florida during the 2020 hurricane season. Data were first used to examine the evacuation and shelter intentions before and during the pandemic. We then measured respondents' hurricane and COVID-19 risk perception, respectively. The impacts of both risk perceptions on respondents' hurricane evacuation intentions were explored. We found that when people felt unsafe to stay home for a Category 2, 3, or 4 hurricane, their intended evacuation was about the same before and during the pandemic regardless of their COVID-19 risk perception. The COVID-19 risk perception, however, significantly lowered the evacuation intention for a Category 1 hurricane. It also significantly influenced evacuees' preference for nontraditional shelters such as government-contracted hotels. The results of our study have practical implications for emergency management and public health governance. Our study also provides insights into decision-making under the conflict between natural hazards and infectious diseases. En este estudio, examinamos la evacuación e intenciones de albergue de familias teniendo en cuenta potenciales conflictos entre los riesgos percibidos de un huracán y las crisis coexistentes en la salud pública. Encuestamos a las familias que residían en las zonas de evacuación por huracanes de la Florida durante la temporada de huracanes del 2020. Los datos se usaron primero para examinar las intenciones de evacuación y de refugio antes y durante la pandemia. Después, medimos la percepción del riesgo de afectaciones por huracanes y COVID-19 de los encuestados, respectivamente. Se exploraron los impactos de ambos tipos de percepciones de riesgo en las intenciones de evacuación, por encuestado. Hallamos que cuando la gente se siente insegura de permanecer en casa frente a huracanes de las categorías 2, 3 y 4, su evacuación intencionada era más o menos la misma de antes y durante la pandemia, sin importar la percepción del riesgo de COVID-19. No obstante, la percepción del riesgo de COVID-19 redujo de manera significativa la evaluación de la intención de evacuación para un huracán de categoría 1. Eso también influyó significativamente en la preferencia de los evacuados por refugios no tradicionales, como los hoteles contratados por el gobierno. ABSTRACT

The growing digital divide issues due to the COVID-19 pandemic are critical to widening disparities and inequalities. Yet, only a few studies have explored the impacts of the digital divide on the education sector. In particular, there is a research gap related to younger students of primary and secondary schools. This study addresses this research gap by providing an overview of the digital divide's impacts on social exclusion and education equality issues. In doing so, the study argues the effects of COVID-19 on the growing digital divide. Such influence is then studied based on the increasing education inequalities due to the digital divide and the eventual social exclusion increase among primary and secondary school children. The study explores the nexus between social exclusion and the digital divide before reflecting on EI as a type of social exclusion. While the main target groups of the study, i.e., primary and secondary school children, are not surveyed, the survey is conducted from two associated stakeholder group, i.e., (1) educators/teachers/managerial members of primary and secondary schools, and (2) parents/guardians of students. A total of 30 schools are surveyed to evaluate the digital divide correlation with "education inequality dimensions" and "social exclusion variables." The vulnerability of schoolchildren from primary and secondary schools is multifaceted as we are yet to witness future psychological and emotional/behavioral impacts of the ongoing pandemic on those affected at a younger age. The paper's findings help verify correlations between DD and social exclusion and education inequality.

ABSTRACT

Upon SARS-CoV-2 infection, viral intermediates specifically activate the IFN response through MDA5-mediated sensing and accordingly induce ADAR1 p150 expression, which might lead to viral A-to-I RNA editing. Here, we developed an RNA virus-specific editing identification pipeline, surveyed 7622 RNA-seq data from diverse types of samples infected with SARS-CoV-2, and constructed an atlas of A-to-I RNA editing sites in SARS-CoV-2. We found that A-to-I editing was dynamically regulated, varied between tissue and cell types, and was correlated with the intensity of innate immune response. On average, 91 editing events were deposited at viral dsRNA intermediates per sample. Moreover, editing hotspots were observed, including recoding sites in the spike gene that affect viral infectivity and antigenicity. Finally, we provided evidence that RNA editing accelerated SARS-CoV-2 evolution in humans during the epidemic. Our study highlights the ability of SARS-CoV-2 to hijack components of the host antiviral machinery to edit its genome and fuel its evolution, and also provides a framework and resource for studying viral RNA editing.

ABSTRACT

The current coronavirus disease 2019 (COVID-19) vaccines are used to prevent viral infection by inducing neutralizing antibody in the body, but according to the existing experience of severe acute respiratory syndrome coronavirus (SARS) infection, T-cell immunity could provide a longer durable protection period than antibody. The research on SARS-CoV-2-specific T-cell epitope can provide target antigen for the development and evaluation of COVID-19 vaccines, which is conducive to obtain COVID-19 vaccine that can provide long-term protection. For screening specific T-cell epitopes, a SARS-CoV-2 S protein peptide library with a peptide length of 15 amino acids was synthesized. Through flow cytometry to detect percentage of IFN-γ+ T cells after mixed COVID-19 convalescent patients' peripheral blood mononuclear cell with peptide library, seven peptides (P77, P14, P24, P38, P48, P74, and P84) that can be recognized by the T cells of COVID-19 convalescent patients were found. After excluding the nonspecific cross-reactions with unexposed population, three SARS-CoV-2-specific T-cell potential epitopes (P38, P48, and P84) were finally screened with the positive reaction rates between 15.4% and 48.0% in COVID-19 convalescent patients. This study also provided the HLA allele information of peptide-positive-response COVID-19 convalescent patients, thus predicting the population coverage of these three potential epitopes. Some HLA alleles showed higher frequency of occurrence in COVID-19 patients than in total Chinese population but no HLA alleles related to the T-cell peptide response and the severity of COVID-19. This research provides three potential T-cell epitopes that are helpful for the design and efficacy evaluation of COVID-19 vaccines. The HLA information provided by this research supplies reference significance for subsequent research such as finding the relation of HLA genotype with disease susceptibility.

ABSTRACT

BACKGROUND: Serological test is helpful in confirming and tracking infectious diseases in large population with the advantage of fast and convenience. Using the specific epitope peptides identified from the whole antigen as the detection antigen is sensitive and relatively economical. The development of epitope peptide-based detection kits for COVID-19 patients requires comprehensive information about epitope peptides. But the data on B cell epitope of SARS-CoV-2 spike protein is still limited. More importantly, there is a lack of serological data on the peptides in the population. In this study, we aimed to identify the B cell epitope peptides of spike protein and detect the reactivity in serum samples, for further providing data support for their subsequent serological applications. RESULTS: Two B cell linear epitopes, P104 and P82, located in non-RBD region of SARS-CoV-2 S protein were identified by indirect ELISA screening of an overlapping peptide library of the S protein with COVID-19 patients' convalescent serum. And the peptides were verified by testing with 165 serum samples. P104 has not been reported previously; P82 is contained in peptide S21P2 reported before. The positive reaction rates of epitope peptides S14P5 and S21P2, the two non-RBD region epitopes identified by Poh et al., and P82 and P104 were 77.0%, 73.9%, 61.2% and 30.3%, respectively, for 165 convalescent sera, including 30 asymptomatic patients. Although P104 had the lowest positive rate for total patients (30.3%), it exhibited slight advantage for detection of asymptomatic infections (36.7%). Combination of epitopes significantly improved the positive reaction rate. Among all combination patterns, (S14P5 + S21P2 + P104) pattern exhibited the highest positive reaction rate for all patients (92.7%), as well as for asymptomatic infections (86.7%), confirming the feasibility of P104 as supplementary antigen for serological detection. In addition, we analyzed the correlation between epitopes with neutralizing antibody, but only S14P5 had a medium positive correlation with neutralizing antibody titre (rs = 0.510, P < 0.01). CONCLUSION: Our research proved that epitopes on non-RBD region are of value in serological detection especially when combination more than one epitope, thus providing serological reaction information about the four epitopes, which has valuable references for their usage.

ABSTRACT

What is already known about this topic? The coronavirus disease 2019 (COVID-19) vaccine development has been progressing, but acceptance of the new vaccines by healthcare workers (HCWs) was not well known prior to approval of COVID-19 vaccines in China. What is added by this report? This study found that before vaccine approval, Beijing HCWs expressed moderate willingness to get vaccinated. Factors positively influencing willingness included free vaccination and belief that the vaccine had been fully evaluated. A negatively influencing factor was presence of an underlying disease. Trust in vaccines, in general, was positively associated with willingness to get new vaccines. What are the implications for public health practice? COVID-19 vaccines should be provided at no cost to HCWs. Effective measures should be taken to enhance the acceptance of COVID-19 vaccination among HCWs in China.

ABSTRACT

PURPOSE: One of the most common types of male infertility is recognized as oligoasthenozoospermia (OA), characterized by low sperm count and quality in males. As a traditional Chinese medicine (TCM), Cuscutae Semen-Mori Fructus coupled-herbs (CSMFCH) has been known to act a curative effect on OA for thousands of years. Nevertheless, the substantial basis and molecular mechanism of CSMFCH in treating OA remain elusive. METHODS: Herein, an integrated approach, including network pharmacology, molecular docking, and experiment validation, was utilized to reveal the new candidate active component and mechanism of CSMFCH in treating OA. RESULTS: The results show that kaempferol is the most significant bioactive component of CSMFCH on OA. The mechanism and targets of CSMFCH against OA are relevant to hormone regulation, oxidant stress, and reproductive promotion. In order to validate network pharmacology results, molecular docking and experiment validation were conducted. In detail, molecular docking was employed to verify the strong binding interactions between kaempferol and the core targets. UHPLC-Q-Orbitrap-MS was used to identify kaempferol in the CSMFCH extract. In vitro and in vivo experiments further proved CSMFCH and kaempferol could enhance the mouse Leydig (TM3) and mouse Sertoli (TM4) cell viability, improve the male reproductive organ weights, sperm quality, and decrease testis tissue damage in the OA mouse model induced by CP. CONCLUSION: Our results not only identify the new candidate active component of CSMFCH in treating OA but also provide new insights into the mechanisms of CSMFCH against OA.

ABSTRACT

Background: Covid-19 vaccine research and development is progressing and expected to be put into use in a predictable time, we aimed to learn the awareness and acceptance of the new vaccine by healthcare workers (HCWs) in Beijing, China. Methods: A cross-sectional survey was conducted to investigate HCWs including doctors, nurses and technicians from sixty hospitals in Beijing to obtain the perception of COVID-19 epidemic and the attitudes towards vaccination before before the completion of vaccine phase III clinical trials. Multivariate analysis was applied to evaluate the associated factors with intention to get vaccination.Results: A total of 8040 HCWs was recruited. 67.1% reported they would get vaccination, others said unsure or would not. Half of the HCWs were unsure whether the outbreak in China would come back and the global epidemic would last for a long time. 67.6% agreed the epidemic can be prevented by vaccination. Positive associated factors with willingness to get vaccination were mainly included epidemic situation prognosis, perception of disease severity, self infection risk and disease can be prevented by vaccine, etc. Two positive factors of “wanted the vaccine free of charge” (OR:5.807, 95%CI:5.083-6.635, P<0.001) and “believed vaccine approved for license have been fully evaluated in clinical trials” (OR:4.485, 95%CI:3.849-5.227, P<0.001) were strongly associated with willingness to get vaccination, while two factors of “highest academic degree” (OR:0.840, 95%CI:0.772-0.914, P<0.001) and “professional ranks and titles” (OR:0.930, 95%CI:0.865-1.000, P=0.049) were negative associated .Conclusions: A little above moderate willingness to get COVID-19 vaccination was found among HCWs in Beijing before the vaccine being licensed. Free vaccination strategy should be considered to implement, effective measures should be taken to remove barriers and convey correct information through appropriate ways to enhance the acceptance of COVID-19 vaccination among HCWs in China.
